FIELD: biotechnology.

SUBSTANCE: vector based on nucleic acids is described used for production of stable cell line packing retrovirus, wherein the vector based on nucleic acids contains the replication beginning site not inherit in mammals and the ability to retain at least 25 thousand pairs of DNA nucleotides (t.p.n.). It is characterized in that the specified vector based on nucleic acids is selected from the bacteria artificial chromosome (BAC), yeast artificial chromosome (YAC), P1-obtained artificial chromosome, fosmid or cosmid. The vector contains retrovirus sequences of nucleic acids encoding gag and pol proteins and env protein, wherein each of retrovirus sequences of nucleic acids has its own promoter inside the vector based on nucleic acids and one or multiple recombination sites for integration of the specified sequences of nucleic acids into endogenous chromosome of the host cell of a mammal in a site-specific way. The invention also relates to applications and methods, in which the specified vector based on nucleic acid is used to produce stable cell lines packing and producing retroviruses.

EFFECT: invention expands the arsenal of means for retrovirus vector production.
