FIELD: biotechnology.

SUBSTANCE: proposed is a method for synthesising a set of oligonucleotides by means of amidophosphite synthesis. The method is implemented on a solid-phase carrier in an apparatus for automatic parallel synthesis. A glass granulate with a diameter of 1 mm is used as an upper filter for reaction cells, after the synthesis of the target fragments, the solid-phase carrier with the attached oligonucleotide fragments is extracted from the reaction cells, mixed in one container with a washing liquid, separated from the granulate, and the remaining mixture is distributed between the cells, the synthesis is continued until the complete sequence of each oligonucleotide from the synthesised set is formed.

EFFECT: invention ensures simultaneous synthesis of a large amount of oligonucleotide primers, a reduction in the amount of the required reaction cells on parallel DNA molecule synthesis apparatuses, the practicability of creating amplicon panels for targeted sequencing with indexing at the stage of the first polymerase chain reaction.
