SUBSTANCE: invention refers to biotechnology and molecular biology. Disclosed is a method of determining a nucleotide target sequence, comprising creating a library for sequencing a new generation, which includes amplification of the target nucleotide sequence using a primer containing a target specific sequence and sequence A, to produce an amplicon, wherein said amplicon can be single-stranded or double-stranded, wherein said sequence A is a known sequence which is identical among the plurality of primers, wherein with all primers containing said sequence A, manipulations and/or amplification can be carried out, identification, sequencing or extraction in the same or similar manner; ligating a first adapter oligonucleotide comprising a B sequence with said amplicon to form an amplicon adapter, wherein said sequence B is a known sequence which is identical among a plurality of primers, wherein with all primers containing said sequence B, manipulations and/or amplification can be carried out, identification, sequencing or extraction in the same or similar manner; and creating a library, which is a "ladder" of fragments containing a plurality of fragments for use as a library for sequencing a new generation; and sequencing fragment of said library, which is a "ladder" of fragments, where the nucleotide sequence determined by sequencing comprises a nucleotide sequence of the target nucleotide sequence.

EFFECT: due to high quality of sequencing with short time of its implementation method can be used in medicine and industry.
