FIELD: biotechnology.
SUBSTANCE: group of inventions is described, including a conjugate of polysaccharide with a protein with high immunogenicity, where the specified conjugate is stable, and it can be used as a vaccine candidate, and a method for preparing the above-mentioned conjugate. In one embodiment, the conjugate contains polysaccharide obtained from gram-negative bacteria, where gram-negative bacterium is Neisseria meningitidis (Men), a carrier protein selected from TT (tetanus toxoid), DT (diphtheria toxoid), CRM197 (non-toxic mutant DT) or OMPV (vesicles from outer membrane proteins), wherein polysaccharide is decomposed in the size range of the distribution coefficient of 0.38±0.06 kD, the conjugate is characterized by a stable carbamate bond -O- (C=O)-N-, and the formula of the specified conjugate is PS-L1-L2-CR, where PS is polysaccharide, L1 is a carbamate bond, L2 is a hydrazide bond, and CR is a carrier protein.
EFFECT: invention expands the arsenal of drugs with high immunogenicity and stability for use as a vaccine candidate.
