FIELD: genetics.
SUBSTANCE: inventions relates to a genetic construct for expression of mNG_CD4-CCR5 genes, a recombinant plasmid and a recombinant vesicular stomatitis virus (VSV) displaying human CD4 and CCR5 receptors on its surface. A genetic construct for the expression of the mNG_CD4 and CCR5 genes is proposed, formed by the sequences of the fluorescent mNeonGreen protein and the human CD4 receptor and co-receptor CCR5 fused through the P2A peptide and having the nucleotide sequence of SEQ ID NO: 1. Proposed recombinant plasmid rVSV_mNeonGreen [CD4-CCR5], encoding human CD4 and CCR5 receptors in the cDNA copy of the genome of the recombinant vesicular stomatitis virus (VSV), having a size of 15259 bp. and containing the above genetic construct. Also proposed is a recombinant VVS strain rVSV_mNeonGreen [CD4-CCR5] containing the above recombinant plasmid and providing virolysis of cells exhibiting HIV-1 gp120/gp41 proteins of R5 tropism on their surface. The strain was deposited in the State collection of causative agents of viral infections, rickettsiosis FBSI SRC VB "Vector" Rospotrebnadzor under the number V-1107.
EFFECT: inventions group provides for the expansion of the range of genetic constructs for the expression of mNG_CD4-CCR5 genes, recombinant plasmids and virus strains that provide virolysis of cells exhibiting gp120/gp41 proteins of HIV-1 tropism R5 on their surface.
