FIELD: biotechnology.
SUBSTANCE: automated method is presented for creating a library of bacterial and yeast cells expressing constructed putative antigens of a T-cell receptor (hereinafter – TCR) on the cell surface. The method includes: obtaining a population of bacterial and yeast cell, processing a cell population on a device for multiplex nuclease-determined genome genetic modification, incubating processed cells for creation of the possibility of genetic modification of nucleic acid in cells, where the genetic modification leads to obtaining nucleic acids encoding constructed peptide antigens in cells, and creating the possibility for cells to express and expose constructed peptide antigens.
EFFECT: invention allows for the most high-performance and reliable identification of specific antigens for TCR in a multiplex way.
