FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, namely to the field of introduction into culture in vitro, production of callus cultures and regenerant plants on solid agar media, using Phlojodicarpus sibiricus (Steph.) K.-Pol as primary explants of vegetative and generative organs. The invention is a method involving the establishment of optimal conditions for the sterilization of primary explants, the determination of the potential for callus formation of various organs and tissues, depending on the type of explants and the culture medium that promotes the induction of callus formation, morphogenesis and organogenesis. The lifetime of calluses, regardless of the primary type of explant, is about 45 days. The color of the callus is different: light yellow, light green, light green with purple inclusions, purple. As the callus dies off, it turns brown. Morphogenic callus cultures are loose, dense, globular in consistency. Organogenesis is observed on Murashige-Skoog’s media with a wide range of concentrations (0.1–2.0 mg/ml) of phytohormones and their combinations (naphthylacetic acid, 2,4-dichlorophenoxyacetic acid, indolylacetic acid, indole-3-butyric acid (IBA), except 6-benzylaminopurine, the addition of which causes the induction of callusogenesis. Rooting of shoots is advisable to carry out using a medium containing IBA, accelerated reproduction of Siberian Alexandrian Laurel under in vitro conditions.
EFFECT: invention makes it possible to use inflorescences, wrapping sheets, and a root as explants, making it possible, regardless of the season of the year, to obtain regenerant plants of a rare medicinal species.
