FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, microbiology and, in particular, to a method for producing latex diagnostic agents for the latex agglutination reaction (LAR). The essence of the invention: the development of a latex diagnostic agent for LAR consists in the fact that polyacrolein latex suspension is centrifuged, adjusted with 0.1 M solution of phosphate-salt buffer (PSB), pH 9.5 to a concentration of 2% on the polymer dry residue and mixed in equal volumes with a water-soluble brucellosis (tularemia) antigen diluted with 0.1 M PSB, pH 9.5 up to a concentration of 1600 mcg/ml, tularemia agent 800 mcg/ml. Immobilization is carried out with stirring and at a temperature of 37°C for 2 hours and 18 hours at a temperature of 4°C, then centrifuged at 5,000 rpm for 10 minutes, washed twice from the unbound antigen 0.1 M PSB, pH 7.2-7.4 and the last precipitate is resuspended in 0.9% sodium chloride solution to 0.2% concentration. Casein up to 0.05% is used to block free diagnostic agent centers. As a preservative, 0.01% sodium merthyolate is used. The setting of the LAR is carried out with the selected diluting liquid Tween 80 in a proportion of 1:50000.
