FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and concerns a method for preparing an erythrocyte immunoglobulin tularemia diagnosticum. The essence of the method is that a 20% suspension of formalinized erythrocytes is obtained for sensitization with tularemia immunoglobulins isolated from immune serum fractionated with caprylic acid. Next, 25 ml of citrate buffer pH-5.0 are added, and incubation for 1 hour at a temperature of 45ºC is carried out. In parallel to 50 ml of immunoglobulins with a protein concentration of 400 mcg/ml on 0.9% sodium chloride solution, 1.5 ml of a conjugating agent - secondary sodium alkyl sulfate at a concentration of 2% is added, and incubation for 1 h at a temperature of 45ºC is carried out. Erythrocytes are mixed on a citrate buffer with conjugated immunoglobulins and incubated for 12 hours at a temperature of 45ºC. The process of double washing from non-bound immunoglobulins is carried out by centrifugation with a 0.1 M solution of phosphate-salt buffer. The process is completed by adding 0.9% sodium chloride solution to a volume of 100 ml and formalin preservative to 1.5%.
EFFECT: use of the method makes it possible to obtain a diagnostic tool for detecting the causative agent of tularemia with high sensitivity and stability.
