METHOD FOR DETERMINATION OF MEMBERS OF FAMILY OF S100 CALCIUM-BINDING PROTEINS BY MEANS OF IMMUNOTURBIDIMETRY Russian patent published in 2022 - IPC G01N33/543 G01N33/68

Abstract RU 2780565 C2

FIELD: biotechnology.

SUBSTANCE: group of inventions relates to turbidimetric immunoassay. A method for in vitro measurement of the presence of calprotectin in a biological sample from a patient, using turbidimetric immunoassay enhanced with latex particles, is disclosed, including stages of collection of predetermined amount of the specified biological sample; extraction of the specified biological sample in predetermined amount of an aqueous organic buffer, and homogenization of a matrix of the specified biological sample, followed by the removal of any material in the form of particles to obtain a sample solution with the presence of solubilized heterodimeric calprotectin; mixing of certain amount of the resulting solution with certain amount of a reagent containing nanoparticles having immobilized two or more monoclonal antibodies or their fragments specifically binding calprotectin to obtain a particle-bound antibody-antigen reaction for calprotectin, which is present as heterodimer S100A8 and S100A9; incubation of the mixture; obtaining optical properties of the mixture, and determination of a signal indicative for calprotectin content based on optical properties of the mixture; establishment of connection of the specified content to the calibrated control, and assessment of a clinical condition of the specified patient based on the measured presence of calprotectin in the specified biological sample. A test kit for implementation of the above-described method is also disclosed.

EFFECT: group of inventions provides the advantage that used reagents create an environment, which is physiologically similar to the environment, into which granulocytes release their physiological calprotectin.
