FIELD: biotechnology and pharmaceutical industry.

SUBSTANCE: invention relates to the field of biotechnology and the pharmaceutical industry and is a method for obtaining total ribonucleic acid (total RNA) from the biomass of the killer yeast Saccharomyces cerevisiae and can be used in pharmaceuticals and biotechnology for the production of anti-infective and immunomodulatory drugs. To implement the method for obtaining total RNA, the yeast biomass is incubated with a buffer solution and a zymolyase enzyme preparation at a temperature of 28-35°C until a homogeneous suspension is obtained, followed by cooling. Next, the resulting suspension is mechanically homogenized under a pressure of 600-1400 bar, centrifuged. Then sodium chloride is added to the resulting homogenate until its concentration reaches 1.5-2.5 M in the target solution, incubated and centrifuged. After that, monohydric alcohol is added to the obtained supernatant to its concentration of 40-60 vol. % in the target solution, the resulting mixture is stirred, incubated and centrifuged to separate the precipitate of total RNA.

EFFECT: present invention allows to increase the yield (approximately 1.3 times) and the purity of total RNA from Saccharomyces cerevisiae yeast cells, as well as the amount of double-stranded RNA in the composition of total RNA.
