METHOD FOR ESTIMATING THE DEGREE OF EXPOSURE OF WORKERS OF PETROCHEMICAL MANUFACTURES TO TOXIC SUBSTANCES Russian patent published in 2022 - IPC G01N33/50 G01N33/68 A61B5/00

Abstract RU 2785267 C1

FIELD: medicine.

SUBSTANCE: invention relates to medicine, namely, to occupational medicine, and can be used to estimate the degree of exposure of workers of petrochemical manufactures to toxic substances, e.g., during a regular medical examination. Peripheral blood mononuclear cells are isolated, applied onto a slide in 1% fusible agarose, lysed in an alkaline solution, subjected to gel electrophoresis, followed by fixing the micro preparations in 70% ethanol for 15 minutes, drying in air at room temperature, and staining in a 0.5% aqueous solution of ethidium bromide. The resulting DNA comets are studied using a fluorescent microscope, the relative content of DNA in the tail of the comets is calculated, after which, for individuals with a percentage of comets with DNA breaks from 0.00 to 19.99%, the degree of exposure of the worker to toxic substances is determined as low; from 20.00 to 39.99%, the degree of exposure to toxic substances is considered average; from 40.00 to 100.00%, a high degree of exposure to toxic substances is determined.

EFFECT: higher accuracy of estimation due to the early detection of DNA breaks.
