FIELD: medicine; diagnosis of parasitic diseases.
SUBSTANCE: invention can be used to determine the viability of Echinococcus. The method is to obtain a monospecific antiserum during surgical intervention in patients with an echinococcal cyst by taking two samples of biomaterial from its lumen, related to viable and non-viable forms of echinococcus, and placing them into 0.9% NaCl. Next, extracts are prepared from the obtained viable and non-viable forms of echinococcus, after which laboratory rabbits are immunized with the obtained extracts. Each rabbit is injected with a mixture containing 0.5 ml of the obtained antigen from the corresponding extract with 0.5 ml of complete Freund's adjuvant. The resulting mixture is injected intradermally into the footpads of all four limbs, subsequent immunization is carried out 8 weeks after the first with the same amount of antigen. On the first day of subsequent immunization, the antigen is administered intramuscularly in the upper thigh, and on the 2nd and 3rd days intravenously in the ear. A week later, immunization is repeated according to the same scheme. The resulting hyperimmune antisera are subjected to the immunosorption procedure to obtain a monospecific antiserum containing a protein antigen of a glycoprotein nature with electrophoretic mobility of alpha 2-globulins, with a molecular weight of 38 kDa according to electrophoresis in 7% PAAG in the presence of sodium dodecyl sulphate, from antibodies obtained from a laboratory rabbit with a viable form, and in the future, with a negative immunological reaction between the obtained antiserum and the blood of a patient with echinococcosis, the echinococcus is determined to be non-viable, and in case of a positive reaction, the echinococcus is determined to be viable.
EFFECT: increased accuracy of diagnosing viable forms of echinococcus in the human body.
