FIELD: biotechnology; virology and medicine.
SUBSTANCE: replication-competent, recombinant oncolytic vaccinia virus (OVV) is proposed. OVV contains a nucleotide sequence encoding an A34 polypeptide variant. The A34 polypeptide variant contains (a) a K151E substitution and (b) an additional substitution selected from the group consisting of the following: R84G, R91A, R91S, F94H и T127E. The following is provided: OVV that contains (a) a nucleotide sequence encoding a B5 polypeptide variant, containing one or more substitutions selected from the group consisting of the following: N39G, L90R, S199M, K229C, S273L, S273V and S273I, and (b) a nucleotide sequence encoding an A34 polypeptide variant that contains the K151E substitution. The following is proposed: OVV that contains (a) a nucleotide sequence encoding an A33 polypeptide variant, containing the A88D and E129M substitutions, and (b) a nucleotide sequence encoding an A34 polypeptide variant, where the A34 polypeptide variant contains the K151E substitution. The A33, A34 and B5 polypeptide variants provide increased viral spread or increased EEV production compared to the corresponding wild-type A33 A34 and B5 polypeptides. The following is proposed: a composition containing OVV for the treatment of cancer and the use of OVV or a composition for the induction of oncolysis in an individual with a tumor.
EFFECT: invention makes it possible to obtain a recombinant oncolytic vaccinia virus with improved antitumor properties.
