FIELD: biotechnology.

SUBSTANCE: method for genetic modification of a primary NK cell is disclosed, including: obtaining a guide RNA (gRNA) specific for a target DNA sequence, and introducing a ribonucleoprotein (RNP) complex containing a Class 2 CRISPR/Cas endonuclease bound into the complex into the target NK cell by electroporation with the corresponding CRISPR/Cas guide RNA hybridizing the target sequence within the genomic DNA of the NK cell. Also a method for the adaptive transfer of engineered NK cells to a subject in need thereof, comprising: obtaining a target NK cell to be modified, obtaining a gRNA specific for the target DNA sequence, introducing an RNP complex into the target NK cell by electroporation, and transferring the engineered NK cells into the subject's body.

EFFECT: effectively used in cancer immunotherapy.
