FIELD: biotechnology; virology; medicine.
SUBSTANCE: arbovirus particle is proposed for generating an immune response to the said arbovirus, comprising a mutant arbovirus that replicates in insect cells and does not replicate in mammalian cells, containing a molecularly altered tetrapeptide furin cleavage site in the precursor polyprotein and optionally a change in the first extension of the upstream amino acids relative to the specified site, and/or a change in the second extension of the amino acids downstream of the specified site. The following is proposed: a composition for developing an immune response to the specified arbovirus, containing an arbovirus particle and a pharmaceutically acceptable carrier, diluent or auxiliary agent; an insect cell including an arbovirus particle for producing said particle; a mammalian cell for inducing an immune response, including a particle of the said arbovirus. The arbovirus carries an altered furin cleavage site that results in increased cleavage of a precursor polyprotein such as prE2 or prM. The virus can be produced in insect’s cells. The virus does not produce viral progeny in mammalian cells.
EFFECT: obtaining mammal specific arbovirus with growth defective.
