SUBSTANCE: invention relates to the field of biotechnology, namely to a nucleic acid construct comprising a sequence of a first polynucleotide comprising a nucleic acid encoding a first DNA-binding protein designed to bind to a defined genomic DNA sequence in a genome, wherein the first DNA-binding protein is a Cas9 protein and a sequence of a second polynucleotide comprising a nucleic acid coding a second DNA-binding protein, which provides for the embedding of the exogenous nucleic acid into the genome, wherein the second DNA-binding protein is a modified hyperactive PiggyBac with improved specificity of embedding the exogenous nucleic acid into the genome as compared to the hyperactive PiggyBac, as well as to a vector, a cell and a composition containing it.
EFFECT: invention is effective for controlled site-specific embedding of one copy or several copies of an exogenous nucleic acid sequence into cells.
