SUBSTANCE: invention can be used to determine the concentration of zinc in the exudate of the “skin window”. A “skin window” method is performed, namely, in the middle third of the palmar surface of the forearm, using a sterile scalpel, the upper layers of the epidermis are removed to the shiny layer, removing the stratum corneum. A plastic chamber with a volume of 1 ml is placed on the scarified area, pre-filled with a syringe with a sterile solution of sodium chloride 0.9%. The camera is fixed to the scarified area of skin using an adhesive tape. After 6 hours, the chamber is removed, its content is collected and transferred to an Eppendorf tube. The exudate of the “skin window” is centrifuged, and the resulting cell-free fraction of the exudate — the supernatant — is immediately used to determine the concentration of zinc using a set of Zinc-Novo reagents from Vector-Best, the main component of which is a specific complexon 2-(5-bromo-2-pyridylazo)-5-(N-propyl-N-sulfopropylamino)-phenol (5-Br-PAPS). The level of zinc is measured on Konelab PRIME 60i automatic biochemical analyzer from Thermo Fisher Scientific; the analyzer automatically processes the sample and, using a built-in photometer with interchangeable interference filters, carries out a photometric measurement of the optical density of the resulting stable complex of the sample at a wavelength of 575 nm.

EFFECT: invention makes it possible to determine in vivo the level of zinc in the exudates of the “skin window” using an accessible and fast method, minimally invasive, in combination with a highly sensitive photocolorimetric method without any deproteinization.
