FIELD: biotechnology.
SUBSTANCE: method of studying the protein composition in biological fluids is described. The method includes mixing a urine sample with a reagent solution, measuring the optical density of the solution photometrically relative to a blank sample, and calculating the protein content from the calibration solution. Quantitative determination of protein concentration involves mixing in a ratio of 1:9 a sample of biological fluid with a complexing reagent containing pyrogallol red 0.05 mmol/L, sodium molybdate 0.16 mmol/L, sodium succinate 50 mmol/L, sodium oxalate 1 mmol/L, 20 vol/vol.%methanol in water, incubating the sample for 10 minutes at room temperature, measuring the optical density of the sample photometrically at a wavelength of 600 and determining the protein concentration.
EFFECT: improvement of analytical characteristics, the sensitivity of the method is 0.05 g/l, the linearity range is up to 2 g/l, the coefficient of variation is less than 10%, the protein yield upon concentration is up to 56–79%.
