FIELD: pharmaceuticals.

SUBSTANCE: invention relates to a method of preparing a compound of formula (A), which includes preparing a compound of formula (A) by reacting a compound of formula (B), a compound of formula (C), a Lewis acid and R5OH, where R5 is an acyl substituted with alkoxy; each of D and D' is independently H or OP1, provided that only one of D and D' is OP1 and A is a group of formula (1), where L is -(CH(OP2))-; R2 is H and P1 is an alkyl or hydroxyl protecting group selected from PNB, or R2 and P1 are combined to form a bond; (i) R3 is H and P2 is a hydroxyl protecting group selected from Bz; (ii) R3 is R-(CH2)nOP5 where P2 is a hydroxyl protecting group selected from PNB and Bz, and P5 is a hydroxyl protecting group selected from PNB and Bz; E is alkyl; G is O; n, if present, is 0, 1, or 2; k is 0 or 1; R1 is -OP6 or -CH2(Y), where P6 is H or a hydroxyl protecting group selected from PNB; R4 is , or where each P7 is a hydroxyl protecting group selected from Pv or Bz; and R6 is alkyl; each Y is -SO2RD; and each RD, if present, is aryl; R7 is either -CHO or where each R7A is independently alkyl. The invention also relates to a method of producing halichondrin macrolide and its analogues and intermediate compounds.

EFFECT: new method of obtaining a compound (A) which can be used for the synthesis of halichondrin macrolide and its analogues.
