FIELD: biotechnology.
SUBSTANCE: invention relates to a method of obtaining a population of immunoreactive T-cells containing a T-cell receptor (TCR) specific for the peptide:MHC complex, where the peptide of the peptide:MHC complex is a mutant GATA3 peptide sequence. The method involves bringing a population of immune cells into contact with a polynucleotide containing a sequence encoding a polypeptide containing the said mutant GATA3 peptide sequence, or a polypeptide containing the said mutant GATA3 peptide sequence. The immune cell population is a population of immune cells lacking cells expressing CD14 and CD25, and wherein the immune cell population includes a population of FMS-like receptor tyrosine kinase ligand 3 (FLT3L) stimulated antigen presenting cells (APCs) and a population of T-cells.
EFFECT: invention is effective for obtaining a population of immunoreactive T-cells for cancer immunotherapy.
