SUBSTANCE: invention relates to a method of purifying a bacterial polysaccharide from a solution containing the said polysaccharide together with contaminants, the said method comprises a flocculation step, which includes the addition of a flocculating agent, and one or more filtration steps, where the flocculating agent contains alum and where the filtration is selected from depth filtration, activated carbon filtration, gel filtration, diafiltration and ultrafiltration, wherein the polysaccharide is a polysaccharide of Streptococcus pneumoniae.
EFFECT: invention provides a simplified purification method that reduces the soluble protein content of bacterial lysates to provide substantially purified bacterial polysaccharides suitable for inclusion in vaccines.
