FIELD: biotechnology.
SUBSTANCE: disclosed is a method for crystallisation and searching for crystallisation conditions of protein molecules, particularly integral membrane proteins, using a lipid sponge phase (LSP) used to create a membrane-like environment of proteins. Method of growing crystals of biological macromolecules comprises steps of preparing a lipid sponge phase, placing a crystallization container in a horizontal position and introduction of a precipitant solution into the working section of the container chamber, introduction of a lipid sponge phase into the working chamber of the container, sealing the inlet of the container and the crystallization container installation on the container holders platform with the possibility of container (1) fixation with the container axis inclination at angle of 0 to 5 degrees relative to platform (7) surface, installation of at least one platform in a thermostatically controlled volume and carrying out incubation for growing protein crystals, opening the inlet of the container with sampling a fraction of the lipid sponge phase with protein crystals and detecting protein crystals. Ratio of the volume of the lipid phase to the volume of the precipitant is selected within range of 1 to 2 to 2 to 1, and the volume of the lipid sponge phase is selected within range of 20 to 80 mcl.
EFFECT: disclosed is a method for crystallisation of membrane proteins in a lipid mesophase for flow crystallography.
