FIELD: biotechnology.
SUBSTANCE: invention relates to immunology, more specifically to genetic modification of immune cells. Presented is a non-replicating recombinant retroviral particle for genetic modification of T- or NK-cells. Particle contains a polynucleotide containing one or more transcription units functionally linked to a promoter active in at least one of the T-cells or NK-cells. One or more transcription units code a first polypeptide containing a constitutively active chimeric lymphoproliferative element. Chimeric lymphoproliferative element includes an extracellular domain which contains a dimerizing motif imparting constitutive activity to the chimeric lymphoproliferative element in the absence of a dimerizing agent. Chimeric lymphoproliferative element is able to stimulate proliferation and/or survival of T-cells or NK-cells and does not contain a cytokine associated with a cognate receptor or a fragment of a cognate receptor.
EFFECT: invention enables to regulate activity of transduced and/or genetically modified T-cells or NK-cells.
