FIELD: biotechnology.
SUBSTANCE: invention is a method for determining pollen viability in vitro in Clarkia Pursh species and varieties, in which the collected pollen of each species or variety is cultivated in two steps, at the same time at the first stage it is sown on a slide in a drop of a liquid medium containing an osmotically active substance – polyethylene glycol with molecular weight 6000 – in concentration of 30% and microelements in the following concentrations – boric acid H3BO3 – 10 mg/100 ml, calcium nitrate CaNO3 – 30 mg/100 ml, magnesium sulphate MgSO4 – 20 mg/100 ml, potassium nitrate KNO3 – 10 mg/100 ml, held for 4 hours at a constant temperature of 25 °C, and at the second stage pollen is kept in this medium for 24 hours, after each stage, energy activity and viability of pollen grains under a microscope and rate of growth of pollen tubes reflecting activity and physiological properties of pollen are monitored.
EFFECT: invention makes it possible to determine viability of pollen in vitro in species and varieties of Clarkia Pursh.
