SUBSTANCE: invention relates to a method of producing a mesenchymal stromal cell (MSC) secretome containing an exosome fraction. Proposed method comprises recovering and culturing mouse adipose tissue-derived MSCs, subsequent detachment of cells using 0.25% trypsin solution and 0.02% Versene solution, three times washing with sterile phosphate buffer solution, centrifugation for 5 minutes at 300 g and room temperature between washes, adding 106 living cells in 500 mcl of phosphate buffer into wells of 24-well plate with incubation at room temperature for 24 hours, after which the supernatant is aspirated, filtered using a syringe filter with pore diameter of 0.2 mcm and centrifuged for 20 minutes at 2,000 g.
EFFECT: invention provides developing a new method of producing MSC secretome containing exosomes of high purity and concentration.
