FIELD: experimental medicine.

SUBSTANCE: disclosed is a method for simulating phosphate calcification on the surface of artificial vessels from an analogue of human blood plasma in vitro, in which solutions of model media are created, which are close in inorganic composition, pH, ionic strength to human blood plasma. To prepare the solution, a solution volume of 250 ml is used, weight (g) of salts: CaCl2·2H2O – 0.1446; K2HPO4 – 0.1288; (NH4)2HPO4 – 0.6600 10−3; MgCl2·6H2O – 0.0482; NaHCO3 – 0.5460; Na2SO4 – 0.0639; NaCl – 5.7330, at pH 7.40±0.05. Plates of artificial vessels are placed in the solution and held for 30–90 days at temperature of 36.6 °C. After that, the plates are removed and dried in air at temperature of 20±5 °C for 5 hours. Composition of the mineral phases and the relative area of the surface of the artificial vessels with calcifications are analyzed.

EFFECT: invention enables to simulate the process of pathogenic crystallisation of carbonate hydroxylapatite with an admixture of whitlockite on the surface of artificial vessels, to create a model system, which can be used to study the efficacy of drugs for preventing and treating pathogenic calcification.
