METHOD FOR PRODUCTION OF COLLAGENASE Russian patent published in 1995 - IPC

FIELD: biotechnology. SUBSTANCE: hepatopancreas of crab is used as raw material for producing collagenase. Isolation is carried out by mixing homogenate of hepatopacreas of crab which is prepared by autolytic hydrolysis and solution of chitosan, the process takes place at pH 2.0-6.5 till final concentration is 0.01-0.4 mass Insoluble material is separated of thus prepared solution. Said solution is affected by ultrafiltration, initially membrane which screens impurities with molecular weight 100 kDa and more is used, then membrane which screens compounds with molecular weight 30 kDa and lower being used. Ferment which does not pass through membrane is collected. Thus prepared ferment solution is lyophilized. Specific activity thus prepared collagenase is 10800-11200 units per 1 g of protein. Mentioned above autolytic hydrolysis is carried out at 0-30 C within 2-8 h with the help of endoprotease. EFFECT: improves efficiency of the method.
