METHOD OF PRODUCING ENZYMATIC COMPLEX PREPARATION HAVING COLLAGENASE ACTIVITY Russian patent published in 2011 - IPC C12N9/48 C12N9/64 

FIELD: chemistry.

SUBSTANCE: method involves formation of a homogenate of a biomass of intestines and heads of larvae of Dermestes frischii or Dermestes maculates beetles in Ringer's solution containing sodium azide and dithiotriethol. Volume ratio of the biomass and Ringer's solution is equal to 1:(0.5-5.0). The supernatant is then separated through decantation of the homogenate. The supernatant undergoes gel filtration on sephadex G-150 with sorption and elution of an enzymatic complex preparation. The obtained preparation undergoes desalination and concentration, after which ion-exchange chromatography with sorption and elution is carried out. The eluted preparation then undergoes desalination and the end product is concentrated. Each desalination process is carried out via column chromatography on sephadex G-25. Each concentration process is carried out through ultra-filtration on membranes with permeability ranging from 5000 to 50000 Da. Output of the obtained preparation is equal to 70% when measuring activity on Z-Ala-Ala-Pro-pNA hydrolysis efficiency and 85% when measuring Mandl activity.

EFFECT: method enables to obtain an enzymatic preparation with high output.
