SUBSTANCE: DNA-fragment cloning encoding a-subunit of leukocyte adhesion receptor LFA-1 is carried out by screening lgt 10 library cDNA prepared from H460 cells induced with complex of phorbol and myristic acid using 32-uniform oligonucleotide 5′-GGGATGTTGTGGTCATGGATGGTGGGCTCAAT as a probe. Clone is isolated hybridizing with oligonucleotide, prepared 5′-Eco RI-fragment of the size 1.0 kb of this clone. cDNA library is subjected for repeated screening using the same probe and clone is isolated that contains additional 5′-fragment of the size 1.0 kb. EFFECT: improved of cloning.
