METHOD OF ASSAY OF MICROBIOLOGICAL WATER POLLUTION Russian patent published in 1997 - IPC

FIELD: medicine, microbiology, ecology. SUBSTANCE: analyzed sample is treated with lysing mixture, DNA-specific fluorescent dye is added and fluorescence intensity is measured at λ (excitement) = 460 ± 5 nm. Microbiological pollution is estimated as epidemiologically hazard if this index exceeds control value (sample of distilled water) by 20%. Water is treated with nonionic detergent in the presence of complexon and at ionic strength corresponding to NaCl concentration above 0.1 M 0.05% solution of Triton X-100 containing 0.1 M Na₂-EDTA and 2 M NaCl at pH = 7.8-8.2 is used as detergent.
