FIELD: biotechnology, biochemistry, medicine. SUBSTANCE: invention relates to preparing highly purified proteolytic enzymes from P. pseudomallei. Method involves the use of two-days broth culture of virulent strain Pseudomonas pseudomallei-57576, protein precipitation at 80% saturation with ammonium sulfate, separation of inert proteins by gel-chromatography on Sephadex G-25 and G-100 and the following purification of enzymes by ion-exchange chromatography on DEAE-Sephadex A-50 at pH = 8.2 and CM-Sephadex C-50 at pH = 5.2. Purification degree is increased by 4-10-fold as compared with prototype. The isolated enzymes can be used for study of melioidosis microbe biology and development of test-system for detection.
