METHOD FOR PREPARING MELIODOSIS PATHOGEN CAPSULE SUBSTANCE Russian patent published in 2004 - IPC

SUBSTANCE: invention proposes a method for preparing capsule substance from meliodosis pathogen. Method involves culturing bacterial mass, extraction of surface cellular biopolymers and fractionation of extracted substance. For increasing capsule formation the normal goat serum is added to nutrient medium in the concentration up to 1.5%. Culturing microorganisms is carried out at 37 C for 24 h followed by centrifugation at 5000g for 10 min. Biopolymers are extracted with solution consisting of 0.3 M of sucrose, 10 mM of tris-HCl, pH 8.0, and 1.5 mM of EDTA. Then supernatant is combined with 5 volumes of cooled acetone, exposed at 0 C for 5 h and centrifuged at 5000g for 10 min. Prepared precipitate is dialyzed against 0.05 M phosphate buffer, pH 7.2, with 0.1 M of NaCl and the prepared substance is subjected for chromatography on matrix TSK HW-60F using 0.05 M phosphate buffer, pH 7.2, with 0.1 M of NaCl as an eluent. The proposed method provides isolating the biopolymer complex as components of B. pseudomallei capsule that is useful for application in immune and immunochemical investigations.
