SUBSTANCE: invention relates to a method of preparing the strain of bacterium assigned for stable storage of plasmid having DNA fragment that provides the betaine utilization from substrate. Method involves carrying out the mutation in chromosomal gene of the strain of bacterium Rhizobium/Agrobacterium that forms deficient strain with respect to betaine utilization. Method involves construction of recombinant plasmid DNA pL 032 containing DNA fragment providing betaine utilization from substrate that has the indicated restriction map. Then recombinant plasmid DNA is transferred in the strain containing mutation followed by culturing the transformed strain of bacterium Rhizobium/Agrobacterium DSM N 6712.
