FIELD: molecular biology, genetic engineering. SUBSTANCE: invention relates to genetic-engineering method of producing S-(+)-2,2-dimethylcyclopropane carboxamide. The latter compound is a parent substance for synthesis of inhibitor of cilastatin dehydropeptidase that is used for prevention of antibiotic inactivation in kidney at effect of renal dehydropeptidase. In medium containing racemic R,S-((±))-2,2- -dimethylcyclopropane carboxamide at pH = 6-11 and 15-55 C a microorganism transformed with recombinant plasmid DNA is added. This DNA fragment encodes an enzyme R-(-)-2,2-dimethyl- -cyclopropanecarboxamide hydrolase of Comamonas or immobilized hydrolase expressed by the indicated microorganism.. The end product is isolated from a mixture containing S-(+)-2,2-dime- -thylcyclopropane carboxamide and R-(-)-2,2-dimethylcyclopropane carboxylic acid. Method is carried out using a new DNA fragment encoding hydrolase of Comamonas, new hybrid recombinant plasmids of Echerichia coli DSM N 7053 and E. coli DSM N 6551 strains transformed with this recombinant DNA plasmid pCA R6. New strain of microorganism-producers are constructed showing increased catalytical and expression capability of gene encoding hydrolase as compared with the known method.
