SUBSTANCE: invention proposes recombinant plasmid DNA containing artificial gene that encodes the fused polypeptide consisting of a single IgG-bindining domain of protein A from Staphylococcus aureus, peptide linker His6-Gly-Ser-Arg and human proinsulin, hybrid tac-promoter and terminator of ribosomal operon of E. coli. Invention proposes the strain of Escherichia coli - producer of fused polypeptide showing the human proinsulin sequence. Biosynthesis of fused polypeptide is induced by isopropylthiogalactopyranoside and level of its biosynthesis is 25% of total cellular protein, not less.
