FIELD: biology. SUBSTANCE: method involves removing hydrocarbon-containing material from cellular nuclei preserved in advance in wheat plantule tissues by means of glycerol medium and then selected and cleaned in glycerol density gradient with following sequential extraction of nuclear fractions of 0.14 M (nucleoplasm), of 0.35 M (chromatin I), and 2 M (chromatin II) with sodium chloride, 6 M guanidine hydrochloride, in the presence of 0.1% β mercaptoethanol (nuclear matrix) by analyzing (determining) hydrocarbon content.
