FIELD: physiology and biochemistry of plants. SUBSTANCE: cell kernels are isolated from tissues of wheal plantules previously preserved in glycerol medium at minus 25 C, and said cell kernels are subjected to stepwise extraction with 0.14M, 0.35M, 2M NaCl, 6M guanidine chloride with 0.1% β-ME and 0.5 NaOH to obtain fractions which are determined for peroxidase activity with benzidine in the presence of H2O2 within 5-10 s from the start of fermentative reaction. The process makes it possible to obtain more accurate and sensitive analysis due to greater recovery of enzyme-containing kernel material and choice of optimum conditions for carrying out chromatic quantitative reaction.
