FIELD: biotechnology. SUBSTANCE: invention relates to preparing derivative of hirudine that acts as a thrombin inhibitor and exhibiting specific activity 10 000 AT U/mg, not less. Amino acid sequence of this derivative of hirudine is identified. Also, nucleotide sequence of recombinant DNA encoding this derivative of hirudine is identified. Method of preparing hirudine derivative involves plasmid construction containing recombinant DNA and its using for transformation of a mutant-producer of Escherichia coli, culturing the transformed cells in medium and isolation of the end product.
