SUBSTANCE: invention relates to 3'-substituted derivative of nucleoside of the following general formula (1) where B means nucleic acid base which can contain a substituent; Z means a lower alkynyl or a lower alkenyl group that can be substituted with a group of the general formula (2) where Ra,Rb and Rc are similar and mean a lower alkyl or phenyl group; R1 and R2 means separately hydrogen atom or benzoyl and R3 means hydrogen atom, residue of mono- or polyphosphoric acid or benzoic acid at condition that a sugar component is ribose, or its pharmaceutically acceptable salt. 3'-Substituted nucleoside derivative shows high antitumor activity and is benefit therefore for treatment and prophylaxis of patients with different cancer species. Invention proposes also methods of synthesis of a compound of the general formula (1) that involves: 1) interaction of sugar derivative of the general formula (3) where R1′,R2′ and R3′ means separately protective groups for hydroxyl-group; Ac means acetyl-group with nucleic acid derivative B-Y where B is indicated above and Y means silyl protective group being a protective group can be removed if necessary; 2) partial hydrolysis of a compound of the formula (5) resulting to formation of a compound of the formula (6) and interaction of formed compound (6) with a compound of the formula Z-X where X means hydrogen or halogen atom or MgBr that results to formation of a compound (7) . The latter is hydrolyzed and then if necessary phosphoric acid ester at one of hydroxyl-group is obtained. Invention relates to also a medicinal composition exhibiting antitumor activity and a method of treatment and prophylaxis of mammals with cancer.
