FIELD: molecular biology, genetic engineering, pharmacy. SUBSTANCE: invention relates to preparing antithrombin polypeptide used for treatment of patients with vein thrombosis. Polypeptides are prepared by genetic engineering methods. Invention involves preparing DNA sequence encoding the end polypeptide precursor bound with leader peptide. DNA sequence is introduced into suitable expression vector containing the gene determining the resistance to antibiotic and bacterial promoter and this vector is used for transformation of cells E. coli type B. Transformed strain is cultured under conditions that are necessary for expression and the end product is isolated. Alternatively, DNA sequence is introduced into transfer vector and insect cell cotransfection is carried out with this vector and DNA baculovirus wild type. Recombinant viral DNA containing DNA sequence that encodes the end polypeptide precursor under polyendrin promoter control is isolated from the transformed cells and this sequence is used for infection of insect cell culture. DNA fragment encoding polypeptide that exhibits an antithrombin activity has the known nucleotide sequence. Recombinant expression and transfer vectors involve this nucleotide sequence. Pharmaceutical composition for treatment of patients with cardiovascular diseases comprises recombinant polypeptide showing an antithrombin activity taken at effective concentration as an active component and a pharmaceutically acceptable carrier or vehicle.
