STRAIN OF BACTERIUM ESCHERICHIA COLI M17 FIM::KAN/P COLAP USED FOR PROBIOTIC PREPARATION PRODUCING Russian patent published in 2000 - IPC

Abstract RU 2144953 C1

FIELD: biotechnology, microbiology. SUBSTANCE: invention relates to preparing producers used for producing probiotic preparations - bacteritic preparations based on live cultures of microorganisms-symbionts used for prophylaxis and treatment of dysbacteriosis and other disorders of gastroenteric tract. Method involves insertion of nonconjugative nonmobilized plasmid pColap carrying genes encoding colicin E1 and factor that determines resistance to ampicillin (doses up to 150 mg/l) into the strain Escherichia coli M17 - producer used for preparing probiotic preparation coli-bacterin. Obtained strain has no nondesirable adhesive phenotype by inactivation of adhesive gene fim H pili of the 1-st type by integration of resistance gene npt to kanamycin-neomycin. The strain E. coli M17 fim H::can/p Colap is deposited in VKPM at N B-7448. Invention ensures to obtain the probiotic preparation showing decreased capacity to colonization and distinct from interstitial niches, increased antagonistic activity and showing residence on the background of therapy with antibiotics carrying out. Traits inserted into the constructed strain are conserved stably. This approach alleviates preparing the standard preparation based on the strain and prevents elimination of plasmid if the preparation is intaken by patient without the accompanying therapy with antibiotics. EFFECT: strain indicated above, valuable properties and traits. 4 dwg, 1 tbl, 3 ex
