METHOD OF DIRECTION OF CELLULAR IMMUNE RESPONSE AGAINST HIV-INFECTED MAMMALIAN CELL, PROTEIN MEMBRANE-BOUND CHIMERIC RECEPTOR, DNA Russian patent published in 2001 - IPC

Abstract RU 2173167 C2

FIELD: medicine, genetic engineering, genetic therapy. SUBSTANCE: method involves the direction of cellular immune response against HIV-infected mammalian cells. Method involves effect of effective dose of cells expressing membrane-bound protein chimeric receptors on HIV-infected cells. Receptor has: (a) extracellular moiety which involves fragment CD4 that is able to recognize and bind HIV-infected cells but it does not mediate HIV infection and (b) intracellular moiety which is able to deliver a signal to indicated cells in order to destruct HIV-infected cells bound with receptor. Protein membrane- bound chimeric receptor has extracellular and intra-cellular components. Extracellular moiety has fragment CD4. Amino acid sequence is given in the invention description. Intracellular moiety is able to deliver a signal to cells carrying the indicated receptor. This signal induces destruction of receptor bound HIV or HIV-infected cells. Intracellular receptor is a moiety of signal-transmitting component of T-cellular receptor, B-cellular receptor or Fc- receptor. Fragment CD4 can be separated from intracellular moiety by transmembrane domain CD7 or CD5, or CD34. Structure of trans-membrane domain is given in the invention description. Invention presents also DNA encoding protein membrane-bound chimeric receptor. Invention provides to make functional chimeras that are able to direct immunocompetent cells for lysis of HIV-infected cells. EFFECT: improved method of direction. 15 cl, 28 dwg, 1 tbl
