METHOD OF PREPARING LEPTOSPIROSIS DIAGNOSTICUM FOR REACTION LATEX-AGGLUTINATION CARRYING OUT Russian patent published in 2001 - IPC

FIELD: medicine, veterinary science, immunology. SUBSTANCE: method involves adsorption of one of components of immune test on surface of latex microcapsules used as carriers and effect of ultrasonic oscillation on products of adsorption in the process of their preparing followed by washing out by centrifugation, incubation in refrigerator, preserving agent addition and bottling. Carboxylated particles activated with carbodiimide are used as latex microspheres and IgG-antibodies to leptospirae of serogroups Canicola and Icterohaemorrhagiae are adsorbed on the above mentioned latex particles by their treatment in ultrasonic bath for 3 ± 0.5 min followed by shaking at 4 ± 1 C and the following incubation of mixture at the same temperature. Then adsorption products are washed out with distilled water, centrifuged and physiological solution is added to obtained precipitate up to initial volume. Method provides early and rapid detection of leptospira antigens and simultaneous identification of their serogroup. Invention can be used for diagnosis of leptospirosis in humans and animals.
