FIELD: medical biotechnology. SUBSTANCE: the innovation deals with the method to test compounds able to induce STF-I transcription. For this purpose the cells of pancreatic insulae which express STF-I/lac Z hybrid gene are isolated, then the impact of different compounds upon STF-I expression is estimated by adding the suggested compound to STF-I/lac Z-expressing cells. Then, due to calorimetric assay a qualitative estimation of lac Z-activity is conducted both in control and treated cells. Due to this method it could be screened the great number of compounds and STF-I- inducing ones could, also, be easily identified. Another aspect of innovation deals with the method to use STF-I promoter to trace insulin-producing cells of pancreatic insulae in vivo. This method uses protein with fluorescence in a green spectrum area as an indicator. Introduction of transgene named "STF-I-protein with green fluorescence" enables to isolate, quickly and efficiently, insulin-producing cells out of pancreas.
