FIELD: biochemistry, molecular biology, genetic engineering. SUBSTANCE: DNA synthesis is carried out using thermostable DNA-polymerase. DNA-polymerase is prepared from Anaerocellum thermophilum or Escherichia coli LE3922pUB520pAR10. DNA-polymerase exhibits 5'-3'-polymerase activity and reverse transcriptase activity but it does not show 3'-5'-exonuclease activity in the presence of magnesium ions and in the absence of manganese ions.
