FIELD: biotechnology, biochemistry, molecular biology, microbiology, amino acids. SUBSTANCE: L-arginine is prepared by culturing microorganism of genus Escherichia transformed with DNA fragment encoding mutant N-acetylglutamate synthase. Mutant N-acetylglutamate synthase is the synthase wherein sequence of amino acids corresponding to positions from 15 to 19 in natural N-acetylglutamate synthase is replaced by any of sequence of amino acids at numbers 1-4 (SEQ ID NOS: 1-4) or the synthase showing indicated properties and containing additionally deletions, substitutions, insertions or additions of one or some amino acids by one multiple positions except for positions from 15 to 19.
