FIELD: biotechnology, microbiology. SUBSTANCE: invention relates to preparing the strain of classic biovar serovar Ogava Vibrio cholerae KM 206 as producer of basic protective antigens using usual media. Method involves conjugated incorporation to cells the natural toxigenic strain Vibrio cholerae Dakka 35 of classic biovar of recombinant plasmid pCT 105 (KmrTcr) with cloned genes of choleraic toxin and the following elimination of plasmid. The level of choleraic toxin in the strain is increased by above 3 times (38.0 mcg/ml) and synthesis of toxin-coregulated adhesion pili is elevated also and protein OmpU appeared. In culturing the strain addition of antibiotics to medium is not required as in the case of plasmid strains. Using this strain allows the enhancement of level of choleraic toxin production with subunit B as the main immunogen and toxin-coregulated adhesion pili also (this causes visible self-agglutination of cells in broth) which is the main colonizing factor of choleraic vibrio. Also, the strain synthesizes protective and immunogenic protein OmpU of external membrane in medium without antibiotics. Invention can be used for preparing vaccines and diagnostic test-systems.
