SUBSTANCE: invention describes set, consisting of complex gene- and immunodiagnostic test-system, which makes it possible to carry out identification of natural strains of cholera vibrion of O1 and O139 serogroups and determine their virulence on the basis of testing presence of diagnostically essential genes and virulence genes and their expression. Gene-diagnostic test-system includes specific primers wbeWl- wbeW2, wbfR1- wbfR2, ctxA1-ctxA2, tcpAc-lassical-tcpAclassica2 and tcpAeltorl-tcpAeltor2 to portions of diagnostically essential genes and virulence genes. Immunoenzymatic test-system on the basis of dot-immunoassay (DIA) contains five nitrocellulose membranes with applied control samples: O1 seroval Inaba antigen, O1 seroval Ogava antigen, O139-antigen, toxin-corregulated adhesion pili, cholera toxin and five polyclonal serums, containing antibodies to said antigens. Immunoenzymatic test-system makes it possible to determine expression of identified genes.
EFFECT: complex geno- and immunodiagnostic test-system has high sensitivity and specificity and makes it possible to carry out multi-factor analysis of natural strains of cholera vibrion, identifying diagnostically important genes and virulence genes, and estimate efficiency of their expression.
