METHOD FOR PREPARING HUMAN HYBRID CELLS EXPRESSING HETEROLOGOUS PROTEIN, HUMAN HYBRID CELLS STRAIN HKB11, STRAIN OF HUMAN HYBRID CELLS IG2 Russian patent published in 2004 - IPC

Abstract RU 2228355 C2

FIELD: biotechnology. SUBSTANCE: invention concerns human hybrid cells. Hybrid cells of human/human are prepared by fusion of human embryo kidney cells (293S) and Burkitt's lymphoma modified cells. Fusion cells can be used as host-cells for recombinant expression of mammal genes. Method provides preparing human hybrid cells strain HKB1, ATCC:CRL-12568 used as a host-cell for expression of heterologous protein eliciting the human profile of glycosylation reaction. Also, method provides preparing human hybrid cells strain IG2, ATCC:PTA-87 expressing protein IL-4SA (T13D/R121E). Advantages in using these hybrid clones of human kidney cells and B- cells named as HKB are the following: cells show negative property by immunoglobulin expression, they show good growth in medium without plasma proteins (with addition of recombinant insulin or without the latter), as suspension cultures in shaker bottle or in fermenter, they show high sensitivity for transfection with DNA and produce abundant amounts of heterologous recombinant proteins, such as recombinant monoclonal antibodies, soluble ICAM-1, rIL-4 and rFVIII. EFFECT: improved preparing method. 15 cl, 3 dwg, 4 tbl, 3 ex
